From where does Inquisit assign the value of its position to on a letter. For example, if the position of "A" is (50,50), does this mean the CENTRE of "A" is in the centre of the screen? Or is it the bottom left or bottom right corner of "A" that's in the 50,50 position/centre of screen?

Yes, that's the default. And you can freely change it via the /hjustify, /vjustify, /halign and /valign attributes.
